Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.
Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
To regain the military initiative, the adoption of mine warfare as an integral strategy of PLAN was discussed at a 1969–70 SWAPO consultative congress held in Tanzania. PLAN's leadership backed the initiative to deploy land mines as a means of compensating for its inferiority in most conventional aspects to the South African security forces. Shortly afterwards, PLAN began acquiring TM-46 mines from the Soviet Union, which were designed for anti-tank purposes, and produced some homemade "box mines" with TNT for anti-personnel use. The mines were strategically placed along roads to hamper police convoys or throw them into disarray prior to an ambush; guerrillas also laid others along their infiltration routes on the long border with Angola. The proliferation of mines in South West Africa initially resulted in heavy police casualties and would become one of the most defining features of PLAN's war effort for the next two decades. On 2 May 1971 a police van struck a mine, most likely a TM-46, in the Caprivi Strip. The resulting explosion blew a crater in the road about two metres in diameter and sent the vehicle airborne, killing two senior police officers and injuring nine others. This was the first mine-related incident recorded on South West African soil. In October 1971, another police vehicle detonated a mine outside Katima Mulilo, wounding four constables. The following day, a fifth constable was mortally injured when he stepped on a second mine laid directly alongside the first.
Dipeptidase 1 (DPEP1), or renal dipeptidase, is a membrane-bound glycoprotein responsible for hydrolyzing dipeptides. It is found in the microsomal fraction of the porcine kidney cortex. It exists as a disulfide-linked homodimer that is glygosylphosphatidylinositol (GPI)-anchored to the renal brush border of the kidney. The active site on each homodimer is made up of a barrel subunit with binuclear zinc ions that are bridged by the Gly125 side-chain located at the bottom of the barrel.

In 1924, Gilbert Walker (for whom the Walker circulation is named) coined the term "Southern Oscillation". He and others (including Norwegian-American meteorologist Jacob Bjerknes) are generally credited with identifying the El Niño effect. The major 1982–83 El Niño led to an upsurge of interest from the scientific community. The period from 1991–1994 was unusual in that El Niños have rarely occurred in such rapid succession. An especially intense El Niño event in 1998 caused an estimated 16% of the world's reef systems to die. The event temporarily warmed air temperature by 1.5 °C, compared to the usual increase of 0.25 °C associated with El Niño events. Since then, mass coral bleaching has become common worldwide, with all regions having suffered "severe bleaching". In June 2026, the NOAA's Climate Prediction Center officially confirmed the return of El Niño, forecasting a strengthening of the phenomenon through the winter of 2026–2027. The agency estimated a 69% likelihood that the event would reach a historically significant intensity, ranking among the strongest El Niño episodes recorded since 1950.
A 2016 paper describes the efforts of how ansuvimab was originally developed as part of research efforts led by Dr. Nancy Sullivan at the United States National Institutes of Health Vaccine Research Center and Dr. J. J. Muyembe-Tamfum from the Institut National de Recherche Biomedicale (INRB) in the Democratic Republic of Congo. This collaborative effort also involved researchers from Institute of Biomedical Research and the United States Army Medical Research Institute of Infectious Diseases. A survivor from the 1995 outbreak of Ebola virus disease in Kikwit, Democratic Republic of Congo donated blood to the project that began roughly ten years after they had recovered. Memory B cells isolated from the survivor's blood were immortalized, cultured and screened for their ability to produce monoclonal antibodies that reacted with the glycoprotein of Ebola virus. Ansuvimab was identified from one of these cultures and the antibody heavy and light chain gene sequences were sequenced from the cells. These sequences were then cloned into recombinant DNA plasmids and purified antibody protein for initial studies was produced in cells derived from HEK 293 cells.

In 2010, she played Tiger Lily in the Pacific Conservatory of the Performing Arts's production of Peter Pan. In 2011, she acted alongside Brendan Cataldo in Leo Cortez's play, Acting Out, at the Alisal Center for the Fine Arts. In 2012, Akagha began playing the leads in various short films and making guest appearances in television series. She appeared in one-off episodes in the series Men at Work, Nashville and 2 Broke Girls. Akagha started voice acting in video games around 2016, first voicing the character "Plastic" in the game Mirror's Edge: Catalyst. In 2017, she was a voice actor for Wolfenstein II: The New Colossus, in which she provided voices for characters Barbara Casey, Zena Woodard, and Angela Cummings. In 2018, she began appearing in Marvel's Runaways television series as Tamar, a recurring, supporting role. She provided the voice for the superheroine Bumblebee in the animated series Teen Titans Go!. Bumblebee initially appeared in the show's fifth season in the episode "Forest Pirates", which aired in 2019. Akagha voiced the main character, Alyx Vance, in the virtual reality game Half-Life: Alyx. The character was first introduced in the game Half-Life 2 in 2004, being originally played by Merle Dandridge. A new actor was chosen for Alyx Vance partially due to the character's younger age and the "time gap" between this and the previous game.
Although the totality of the data that have been reviewed provides reassurance, pancreatitis will continue to be considered a risk associated with these drugs until more data are available; both agencies continue to investigate this safety signal." Lawsuits have been filed in which plaintiffs who developed pancreatic cancer claim that DPP-IV inhibitors or incretins had a causative role in the development of their cancers.
had reported in February 2015 that Tagliavini was able to convince pro-Russian separatists during the negotiations of the Minsk II agreement because she had the benefit of Putin's trust. Saakashvili said in an interview in August 2019: "It was a politicised commission. It was quite bad because the whole idea behind the commission was to get the responsibility off Russia, to share the blame. [...] The Americans were categorically against it. [...] it was a Steinmeier game; I think backed by Merkel to dilute Russia's blame." The Atlantic Council members stated on anniversary of the war in 2021 that Russia and South Ossetia initiated the 2008 conflict and that the EU report was erroneous.
Six different types of vancomycin resistance are shown by enterococcus: Van-A, Van-B, Van-C, Van-D, Van-E and Van-G. The significance is that Van-A VRE is resistant to both vancomycin and teicoplanin, Van-B VRE is resistant to vancomycin but susceptible to teicoplanin, and Van-C is only partly resistant to vancomycin. The mechanism of resistance to vancomycin found in Enterococcus involves the alteration of the peptidoglycan synthesis pathway. The D-alanyl-D-lactate variation results in the loss of one hydrogen-bonding interaction (four, as opposed to five for D-alanyl-D-alanine) being possible between vancomycin and the peptide. The D-alanyl-D-serine variation causes a six-fold loss of affinity between vancomycin and the peptide, likely due to steric hindrance.
A peptide microarray (also commonly known as peptide chip or peptide epitope microarray) is a collection of peptides displayed on a solid surface, usually a glass or plastic chip. Peptide chips are used by scientists in biology, medicine and pharmacology to study binding properties and functionality and kinetics of protein-protein interactions in general. In basic research, peptide microarrays are often used to profile an enzyme (like kinase, phosphatase, protease, acetyltransferase, histone deacetylase etc.), to map an antibody epitope or to find key residues for protein binding. Practical applications are seromarker discovery, profiling of changing humoral immune responses of individual patients during disease progression, monitoring of therapeutic interventions, patient stratification and development of diagnostic tools and vaccines.

== Treatment == Treatment is indicated when the so-called table-top test is positive. With this test, the person places their hand on a table. If the hand lies completely flat on the table, the test is considered negative. If the hand cannot be placed completely flat on the table, leaving a space between the table and a part of the hand as big as the diameter of a ballpoint pen, the test is considered positive and surgery or other treatment may be indicated. Additionally, finger joints may become fixed and rigid. There are several types of treatment, with some hands needing repeated treatment. The main categories listed by the International Dupuytren Society in order of stage of disease are radiation therapy, needle aponeurotomy (NA), collagenase injection, and hand surgery. As of 2016 the evidence on the efficacy of radiation therapy was considered inadequate in quantity and quality, and difficult to interpret because of uncertainty about the natural history of Dupuytren's disease. Needle aponeurotomy is most effective for Stages I and II, covering 6–90 degrees of deformation of the finger. However, it is also used at other stages. Collagenase injection is likewise most effective for Stages I and II. However, it is also used at other stages. Hand surgery is effective at stage I to stage IV. Use of a splint to keep treated fingers straight following various forms of treatment, typically at all times for some days, then at nighttime for some weeks, is usual.
Until 2009 all species within the family Conidae were placed in one genus, Conus. Testing of the molecular phylogeny of the Conidae was first conducted by Christopher Meyer and Alan Kohn, and has continued, particularly with the advent of nuclear DNA testing. In 2009, J.K. Tucker and M.J. Tenorio proposed a classification system consisting of three distinct families and 82 genera for living species of cone snails. This classification is based on shell morphology, radular differences, anatomy, physiology, and cladistics, with comparisons to molecular (DNA) studies. Published accounts of Conidae that use these new genera include J.K. Tucker & M.J. Tenorio (2009), and Bouchet et al. (2011). Tucker and Tenorio's proposed classification system for the cone shells and other clades of Conoidean gastropods is shown in Tucker & Tenorio cone snail taxonomy 2009. Some experts, however, still prefer to use the traditional classification. For example, in the November 2011 version of the World Register of Marine Species, all species within the family Conidae were placed in the genus Conus. The binomial names of species in the 82 genera of living cone snails listed in Tucker & Tenorio 2009 were recognized by the World Register of Marine Species as "alternative representations". Debate within the scientific community regarding this issue has continued, and additional molecular phylogeny studies are being carried out in an attempt to clarify the issue.
Moscow Mayor Sergei Sobyanin announced that the newly opened Moscow-based "R-Pharm" will become a leading manufacturer of Russia's Sputnik V coronavirus vaccine. Working at full capacity, the factory will produce up to 10 million doses a month. In May 2021 Sergei Sobyanin complained that only 1.3 million Moscow residents out of 12 million had received the first dose (10.2%). Only 9.5% of Russians had received a vaccine. Forbes Russia established that Russia committed to export 205 millions of doses of "Sputnik V" to other countries, and as of 16 May.3 millions (8%) were so far delivered. A survey found that 62% of the Russian population felt hesitant, with 55% not afraid of getting sick and some willing to wait for CoviVac. In June 2021, with the increase in Delta variant cases, several Russian city governments introduced strict measures to overcome vaccine hesitancy, such as requiring vaccine QR codes from customers in cafes.
