Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.
Stability studies focus on deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation into higher-order species. The fatty acid side chain adds susceptibility to oxidative change and can promote self-association at high concentration. Lyophilised material is comparatively robust when kept cold and dry, while aqueous solutions require refrigeration and protection from light. Forced degradation experiments under heat, acid, base, and peroxide conditions establish the specificity of each analytical method. Which degradation route dominates under real storage conditions depends on the formulation and stays formulation-specific.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.
Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.

Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
=== Notable people === The American pop singer Michael Jackson had the condition, which he obscured with costumes and makeup until it covered his body. This led to some inaccurate speculation that he had bleached his skin. Other notable cases include the American rapper Krizz Kaliko, the Canadian fashion model Winnie Harlow, the New Zealand singer-songwriter Kimbra, the American actor David Dastmalchian, the Argentine musician Charly García, the professional wrestler Bryan Danielson, the French actor Michaël Youn, the former French Prime Minister Édouard Philippe, Miss Universe Egypt 2024 Logina Salah, governor of Pampanga Eddie Panlilio, and the Colombian model Taliana Vargas.
=== Interleukins and obesity === With the discovery of interleukins (IL), the concept of systemic inflammation developed. Although the processes involved are identical to tissue inflammation, systemic inflammation is not confined to a particular tissue but involves the endothelium and other organ systems. Chronic inflammation is widely observed in obesity. Obese people commonly have many elevated markers of inflammation, including:
=== Active site === The cleft between GALE's N- and C-terminal domains constitutes the enzyme's active site. A conserved Tyr-X-X-X Lys motif is necessary for GALE catalytic activity; in humans, this motif is represented by Tyr 157-Gly-Lys-Ser-Lys 161, while E. coli GALE contains Tyr 149-Gly-Lys-Ser-Lys 153. The size and shape of GALE's active site varies across species, allowing for variable GALE substrate specificity. Additionally, the conformation of the active site within a species-specific GALE is malleable; for instance, a bulky UDP-GlcNAc 2' N-acetyl group is accommodated within the human GALE active site by the rotation of the Asn 207 carboxamide side chain.
The oldest and most widely used expression systems are cell-based and may be defined as the "combination of an expression vector, its cloned DNA, and the host for the vector that provide a context to allow foreign gene function in a host cell, that is, produce proteins at a high level". Overexpression is an abnormally and excessively high level of gene expression which produces a pronounced gene-related phenotype. There are many ways to introduce foreign DNA to a cell for expression, and many different host cells may be used for expression — each expression system has distinct advantages and liabilities. Expression systems are normally referred to by the host and the DNA source or the delivery mechanism for the genetic material. For example, common hosts are bacteria (such as E. coli, B. subtilis), yeast (such as S. cerevisiae) or eukaryotic cell lines. Common DNA sources and delivery mechanisms are viruses (such as baculovirus, retrovirus, adenovirus), plasmids, artificial chromosomes and bacteriophage (such as lambda). The best expression system depends on the gene involved, for example the Saccharomyces cerevisiae is often preferred for proteins that require significant posttranslational modification. Insect or mammal cell lines are used when human-like splicing of mRNA is required. Nonetheless, bacterial expression has the advantage of easily producing large amounts of protein, which is required for X-ray crystallography or nuclear magnetic resonance experiments for structure determination.

The US Federal Aviation Administration issued a notice to airmen on 3 January 2026 prohibiting US aircraft from operating within Venezuelan airspace, citing "ongoing military activity". The US Embassy in Caracas issued a shelter-in-place order from its relocated operations in Colombia. The southern area of Caracas reportedly lost power. Republican Senator Mike Lee said that Secretary of State Marco Rubio "anticipates no further action in Venezuela now that Maduro is in US custody," and that Maduro was expected to go on trial in the US. In the hours after the US attack, Rubio placed a call with Delcy and Jorge Rodríguez, as well as Mauricio Claver-Carone, who was involved in the plan to abduct Maduro and would go on to serve as an unofficial viceroy for the Trump administration in Venezuela. In a television interview with ABC News, Rubio thanked news organizations who had received leaks about the operation before it took place for not reporting on it. According to Semafor, The New York Times and The Washington Post had received information about the attack but did not report on it in order to not endanger the lives of US personnel involved in the operation. The acknowledgment of the media withholding information about the operation came after defense secretary Pete Hegseth imposed strict restrictions on Pentagon reporters due to a mistrust of journalists' abilities to withhold militarily sensitive information from the public.
=== Cell Biology and Virology === Organised into subprograms including Eukaryotic Microbiology, Tumor Cell Biology, Virology, and Mammalian Cell Structure/Differentiation, this program studies cellular functions across organisms. Research areas include tumor-virus associations, host-pathogen signaling, gene therapy, and vaccine development.
In 2013, between 128 and 232 million people used cannabis (2.7% to 4.9% of the global population between the ages of 15 and 65). Cannabis is by far the most widely used illicit substance, with the highest use among adults (as of 2018) in Zambia, the United States, Canada, and Nigeria.
=== Basal metabolic rate === Basal metabolic rate (BMR) is one of the main components of a person's daily energy expenditure. It is defined as the amount of energy that is expended during a given amount of time by a person at rest. This energy at rest is used to pump blood throughout the body, maintain proper brain function, break down toxins, and ensure other bodily functions. Technically speaking, BMR is the energy the body expends during the following specific conditions: immediately after waking up, while in a resting state, and after fasting for 12–14 hours. Sometimes the term Resting Metabolic Rate (RMR) is used in place of BMR, but RMR is not solely measured under the previously listed stringent conditions and it is about 10% more than BMR. The BMR is directly proportional to a person's lean body mass. In other words, the more lean body mass a person has, the higher their BMR. BMR is also affected by acute illnesses and increases with burns, fractures, infections, fevers, etc. It can be measured via direct and indirect calorimetry. However, it is also possible to approximately estimate BMR using several equations that factor in a person's age, sex, height, and weight. Some of the most popular and accurate equations used to calculate BMR are the original Harris-Benedict equations, the revised Harris-Benedict equations, and the Mifflin St. Jeor equation. The original Harris-Benedict Equations are as follows:
AgNPs are synthesized using microwave irradiation, gamma irradiation UV activation, or conventional heating of the precursor silver nitrate, AgNO3 using an alginate solution as a stabilizing and reducing agent. The carboxyl or hydroxyl groups on the alginate reagent form complexes during the synthesis of the AgNPs that stabilize the reaction. Nanoparticle size and shape can be specified by changing the ratio of alginate to silver nitrate used and/or the pH. A coating such as PVP may be added to the nanoparticles by heating and subsequent slow cooling.

Methods have also advanced dramatically, advancing from examination of animals through dissection of fresh and preserved cadavers (corpses) to technologically complex techniques developed in the 20th century.
==== Membrane filtration ==== Membrane filtration can remove suspended solids and organic components, and inorganic pollutants such heavy metals. For heavy metal removal, several forms of membrane filtration, such as ultrafiltration, nanofiltration, and reverse osmosis, can be used depending on the particle size that can be maintained. Antiscalants can help maintain membrane filtration. Some small molecules can permeate to some extent through membranes.

Purity is commonly expressed as the percentage of the main peak relative to all integrated peaks in a reversed-phase chromatogram. Related substances and counter-ions are reported separately. Values obtained with different detectors are not always directly comparable.
Aggregation is driven by hydrophobic contacts, especially those involving the fatty acid side chain, and is accelerated by heat, agitation, and repeated freezing and thawing. Low pH and suitable excipients can reduce the rate. The tendency differs between formulations.
Dry powder has low molecular mobility and tolerates colder storage for longer periods. Water enables hydrolysis and conformational change, so dissolved material is kept refrigerated and used within a shorter window.
Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and cleared quickly. Semaglutide carries a position 8 substitution that blocks that cleavage and a fatty diacid chain that binds albumin. Together these changes extend its circulating half-life to about one week.
