Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.

Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
=== Retro-Diels-Alder reaction === This reaction occurs mainly in cyclohexene and its derivatives. Upon ionization, the pi electrons are excited and generate a charge site and a radical site. Following this, two successive α cleavages yield a butadiene radical and a neutral ethene since ethene has a higher ionisation energy than butadiene ( Stevenson's rules).
2,5-DMA has been encountered as a novel designer drug, with capsules of 200 mg of the hydrobromide salt, equivalent to 170 mg of the hydrochloride salt. It was initially misrepresented as mescaline or MDA before being named DMA. According to Shulgin, this might be an effective dose level that has not been formally clinically explored.
=== Pharmacoepidemiology and therapeutics === A 2023 study in JAMA Network Open examined changes in prescribing patterns of oral minoxidil for hair loss after media attention, using prescription data from U.S. health-system records, including those affiliated with Truveta. A 2025 study, also in JAMA Network Open, examined patterns of discontinuation and reinitiation of dual-labeled GLP-1 receptor agonists among U.S. adults with overweight or obesity using Truveta data. In 2024, a study in JAMA Internal Medicine compared the effectiveness of semaglutide versus tirzepatide for weight loss in adults with overweight or obesity, using data from Truveta-affiliated health systems.
Somatostatin receptor antagonists (or somatostatin inhibitors) are a class of chemical compounds that work by imitating the structure of the neuropeptide somatostatin, which is an endogenous hormone found in the human body. The somatostatin receptors are G protein-coupled receptors. Somatostatin receptor subtypes in humans include sstr1, 2A, 2 B, 3, 4, and 5. While normally expressed in the gastrointestinal (GI) tract, pancreas, hypothalamus, and central nervous system (CNS), they are expressed in different types of tumours. The predominant subtype in cancer cells is the ssrt2 subtype, which is expressed in neuroblastomas, meningiomas, medulloblastomas, breast carcinomas, lymphomas, renal cell carcinomas, paragangliomas, small cell lung carcinomas, and hepatocellular carcinomas. As a radiopharmaceutical compound that is selective for somatostatin receptors, there is research being done for these radiolabeled compounds to act as diagnostic tests in PET scans for neuroendocrine tumors and other tumors not previously targeted with radiolabeled somatostatin receptor agonists, and to act as radiopharmaceutical therapeutic compound, more specifically to conduct peptide radionuclide receptor therapy. Some non-radiopharmaceutical compounds that are developed as competitive inhibitors of somatostatin, such as the hormone antagonist cyclosomatostatin.

The hormonal IUD is a small T-shaped piece of plastic, which contains levonorgestrel, a type of progestin. The cylinder of the device is coated with a membrane that regulates the release of the drug. Bayer markets Skyla as Jaydess in the United Kingdom. Jaydess releases six micrograms per day and lasts for three years. In comparison, oral contraceptives can contain 150 micrograms of levonorgestrel. The hormonal IUD releases the levonorgestrel directly into the uterus, as such its effects are mostly paracrine rather than systemic. Most of the drug stays inside the uterus, and only a small amount is absorbed into the rest of the body.
=== Diaphragm check valve === A diaphragm check valve uses a flexing rubber diaphragm positioned to create a normally-closed valve. Pressure on the upstream side must be greater than the pressure on the downstream side by a certain amount, known as the pressure differential, for the check valve to open allowing flow. Once positive pressure stops, the diaphragm automatically flexes back to its original closed position. This type is used in respirators (face masks) with an exhalation valve.
== Corneal keratocytes == Keratocytes are flattened cells found dispersed within the corneal stroma. The primary role of this sparse population of cells is thought to be in maintaining the extracellular matrix of collagen lamellae that surround them. However, keratocytes also play a defensive role during pathogenic invasion. They can be influenced by IL-1α (secreted by corneal epithelial cells) and tumor necrosis factor (TNF)-α to produce both IL-6 and defensins. Of these, the former is found to combine synergistically with other interleukins to increase co-stimulation of other immune aspects as well as increase antibody secretion. The latter, defensins, have a wide range of antimicrobial affects against bacteria, fungi, and viruses, as well as effects in accelerating healing of damaged epithelial cells. It has also been found that the presence of secreted defensins secreted by corneal keratocytes is correlated with cases of corneal transplant rejection, suggesting that these peptides may have a role in tissue rejection. Furthermore, keratocytes have also been found to secrete IL-8, which attracts neutrophils, in infections involving the herpes simplex virus.
Most bark is collected illegally by local people who are paid 150 CFA francs per kilo (about US$0.10 per pound) for delivery of pre-dried bark at the roadside. In practice they confuse and mix it with P. macroceras ("false yohimbe"), a species that contains little yohimbine.
The provisions of the Codex Alimentarius are that any "first generation" product must be labeled "irradiated" as any product derived directly from an irradiated raw material; for ingredients the provision is that even the last molecule of an irradiated ingredient must be listed with the ingredients even in cases where the unirradiated ingredient does not appear on the label. The RADURA-logo is optional; several countries use a graphical version that differs from the Codex-version. The suggested rules for labeling is published at CODEX-STAN – 1 (2005), and includes the usage of the Radura symbol for all products that contain irradiated foods. The Radura symbol is not a designator of quality. The amount of pathogens remaining is based upon dose and the original content and the dose applied can vary on a product by product basis. The European Union follows the Codex's provision to label irradiated ingredients down to the last molecule of irradiated food. The European Union does not provide for the use of the Radura logo and relies exclusively on labeling by the appropriate phrases in the respective languages of the Member States. The European Union enforces its irradiation labeling laws by requiring its member countries to perform tests on a cross section of food items in the market-place and to report to the European Commission. The results are published annually on EUR-Lex. The US defines irradiated foods as foods in which the irradiation causes a material change in the food, or a material change in the consequences that may result from the use of the food.

Nicotinamide riboside kinase 1 is an enzyme that in humans is encoded by the NMRK1 gene. As both a nicotinate riboside kinase (EC 2.7.1.173), and a ribosylnicotinamide kinase (EC 2.7.1.22), this enzyme helps perform a step in the process of converting certain forms of niacin/Vitamin B3 into active coenzymes like NAD+. In humans, nicotinamide riboside kinase 1 is expressed in most tissues of the body, compared to the other isozyme, nicotinamide riboside kinase 2, which is predominantly found in muscles.
== SUMO attachment (SUMOylation) == SUMO attachment to its target is similar to that of ubiquitin (as it is for the other ubiquitin-like proteins such as NEDD 8). The SUMO precursor has some extra amino acids that need to be removed, therefore a C-terminal peptide is cleaved from the SUMO precursor by a protease (in human these are the SENP proteases or Ulp1 in yeast) to reveal a di-glycine motif. The obtained SUMO then becomes bound to an E1 enzyme (SUMO Activating Enzyme (SAE)) which is a heterodimer (subunits SAE1 and SAE2). It is then passed to an E2, which is a conjugating enzyme (Ubc9). Finally, one of a small number of E3 ligating proteins attaches it to the protein. In budding yeast, there are four SUMO E3 proteins, Cst9, Mms21, Siz1 and Siz2. While in ubiquitination an E3 is essential to add ubiquitin to its target, evidence suggests that the E2 is sufficient in SUMOylation as long as the consensus sequence is present. It is thought that the E3 ligase promotes the efficiency of SUMOylation and in some cases has been shown to direct SUMO conjugation onto non-consensus motifs. E3 enzymes can be largely classed into PIAS proteins, such as Mms21 (a member of the Smc5/6 complex) and Pias-gamma and HECT proteins. On Chromosome 17 of the human genome, SUMO2 is near SUMO1+E1/E2 and SUMO2+E1/E2, among various others. Some E3's, such as RanBP2, however, are neither. Recent evidence has shown that PIAS-gamma is required for the SUMOylation of the transcription factor yy1 but it is independent of the zinc-RING finger (identified as the functional domain of the E3 ligases).
== Complete and incomplete separation == Some types of separation require complete purification of a certain component. An example is the production of aluminum metal from bauxite ore through electrolysis refining. In contrast, an incomplete separation process may specify an output to consist of a mixture instead of a single pure component. A good example of an incomplete separation technique is oil refining. Crude oil occurs naturally as a mixture of various hydrocarbons and impurities. The refining process splits this mixture into other, more valuable mixtures such as natural gas, gasoline and chemical feedstocks, none of which are pure substances, but each of which must be separated from the raw crude. In both complete separation and incomplete separation, a series or cascade of separations may be necessary to obtain the desired end products. In the case of oil refining, crude is subjected to a long series of individual distillation steps, each of which produces a different product or intermediate.

It is a synthetic analog of GLP-1 produced through medicinal chemistry to resist enzymatic degradation. The design goal was longer circulation than the native hormone.
A fatty acid side chain attaches the peptide to serum albumin, which shields it from kidney filtration and protease activity. This interaction is the main reason its circulation time is extended.
No. The native hormone is GLP-1, and semaglutide is an engineered variant with three deliberate structural alterations. It does not appear in unmodified biological sources.
Lower temperatures slow hydrolysis, oxidation and aggregation reactions that degrade the molecule over time. Lyophilised powder is more tolerant than solution, but both benefit from controlled conditions.
