GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
===== Activated carbon ===== Activated carbons (ACs) or biological-activated carbon (BAC) are effective adsorbents for a wide variety of contaminants. The adsorptive removal of color, aroma, taste, and other harmful organics and inorganics from drinking water and wastewater is one of their industrial applications. Both a high surface area and a large pore size can improve the efficiency of activated carbon. Activated carbon was utilized by a number of studies to remove heavy metals and other types of contaminants from wastewater. The cost of activated carbon is rising due to a shortage of commercial activated carbon (AC). Because of its high surface area, porosity, and flexibility, activated carbon has a lot of potential in wastewater treatment.
== Music == The origins of music during the Paleolithic are unknown. The earliest forms of music probably did not use musical instruments other than the human voice or natural objects such as rocks. This early music would not have left an archaeological footprint. Music may have developed from rhythmic sounds produced by daily chores, for example, cracking open nuts with stones. Maintaining a rhythm while working may have helped people to become more efficient at daily activities. An alternative theory originally proposed by Charles Darwin explains that music may have begun as a hominin mating strategy. Bird and other animal species produce music such as calls to attract mates. This hypothesis is generally less accepted than the previous hypothesis, but nonetheless provides a possible alternative. Upper Paleolithic (and possibly Middle Paleolithic) humans used flute-like bone pipes as musical instruments, and music may have played a large role in the religious lives of Upper Paleolithic hunter-gatherers. As with modern hunter-gatherer societies, music may have been used in ritual or to help induce trances. In particular, it appears that animal skin drums may have been used in religious events by Upper Paleolithic shamans, as shown by the remains of drum-like instruments from some Upper Paleolithic graves of shamans and the ethnographic record of contemporary hunter-gatherer shamanic and ritual practices.
The oldest known constructed roadways are the stone-paved streets of the city-state of Ur, dating to c. 4,000 BCE, and timber roads leading through the swamps of Glastonbury, England, dating to around the same period. The first long-distance road, which came into use around 3,500 BCE, spanned 2,400 km from the Persian Gulf to the Mediterranean Sea, but was not paved and was only partially maintained. Around 2,000 BCE, the Minoans on the Greek island of Crete built a 50 km road leading from the palace of Gortyn on the south side of the island, through the mountains, to the Palace of Knossos on the north side of the island. Unlike the earlier road, the Minoan road was completely paved. Ancient Minoan private homes had running water. A bathtub virtually identical to modern ones was unearthed at the Palace of Knossos. Several Minoan private homes also had toilets, which could be flushed by pouring water down the drain. The ancient Romans had many public flush toilets, which emptied into an extensive sewage system. The primary sewer in Rome was the Cloaca Maxima; construction began on it in the sixth century BCE, and it is still in use today. The ancient Romans also had a complex system of aqueducts, which were used to transport water across long distances. The first Roman aqueduct was built in 312 BCE. The eleventh and final ancient Roman aqueduct was built in 226 CE. Put together, the Roman aqueducts extended over 450 km, but less than 70 km of this was above ground and supported by arches.
Thursday of the Dead is a feast day shared by Christians and Druze in the Lebanon, it falls sometime between the Easter Sundays of the Catholic and Eastern Orthodox Christian traditions. It is a day on which the souls of the dead are honoured. Particularly popular among women in the region, this occasion highlights the shared cultural heritage between Arab Christians and Druze in Lebanon. Additionally, the baptism of children in accordance with Christian customs often took place within prominent Lebanese Druze families. Common surnames among Lebanese Christians and Druze include Abi-Lamma, Assaf, Atrash, Awar, Ballout, Barakat, Daou, Dergham, Faour, Farraj, Ghannam, Halabi, Harb, Hatoum, Hilal, Ibrahim, Jaber, Kadamani, Kadi, Malaeb, Nammour, Safadi, Saker, Saleh, Serhal, Shaya, Timani, Yaghi, and others.

After rail services were extended to Dover in 1844, the Western Docks were used as a terminal for the Golden Arrow and other cross-channel train services with its own railway station, Dover Marine, later renamed Dover Western Docks. At Dover Marine station an estimated 5 million troops departed for the trenches of World War I and nearly 1.5 million wounded soldiers returned. In 1920 the remains of the Unknown Warrior were landed prior to transportation to London and a ceremonial interment at Westminster Abbey. Dover Marine railway station closed in 1994, later to be redeveloped into Cruise Terminal One. The Western Docks area was also used from 1968 to the early 2000s for a cross-channel hovercraft service run by Hoverspeed. Hoverspeed also ran catamaran services until being declared bankrupt in 2005. Another catamaran service was run from 2004 until November 2008 by the single ship of SpeedFerries, SpeedOne, with up to five services daily to Boulogne-sur-Mer. The hoverport has now been demolished and redeveloped as a cargo handling facility.
Three of the four outer-shell electrons of each atom in a graphene sheet occupy three sp2 hybrid orbitals – a combination of orbitals s, px and py — that are shared with the three nearest atoms, forming σ-bonds. The length of these bonds is about 0.142 nanometers. The remaining outer-shell electron occupies a pz orbital that is oriented perpendicularly to the plane. These orbitals hybridize together to form two half-filled bands of free-moving electrons, π, and π∗, which are responsible for most of graphene's notable electronic properties. Recent quantitative estimates of aromatic stabilization and limiting size derived from the enthalpies of hydrogenation (ΔHhydro) agree well with the literature reports. Graphene sheets stack to form graphite with an interplanar spacing of 0.335 nm (3.35 Å). Graphene sheets in solid form usually show evidence in diffraction for graphite's (002) layering. This is true of some single-walled nanostructures. However, unlayered graphene displaying only (hk0) rings have been observed in the core of presolar graphite onions. TEM studies show faceting at defects in flat graphene sheets and suggest a role for two-dimensional crystallization from a melt.
=== Resistant gram negative bacteria === Investigators in the National Human Genome Research Institute and the NIH Clinical Center have used advanced DNA sequencing methods to characterize carbapenem-resistant enterobacteriaceae in hospitalized patients. They tracked a cluster of infections in hospitalized patients, then more recently, Frank's laboratory conducted a two-year follow-up study to understand the possible spread of the resistant genes between bacteria on plasmids. To further investigate the mechanisms used by bacteria to spread resistance, Frank's research program examined the frequency of horizontal gene transfer among species, using bacterial isolates from two hospitals. Most conjugation studies use a lab strain of E. coli, typically under narrow conditions. Given that conjugation is influenced by multiple factors, we sought to perform a systematic analysis of blaKPC encoding plasmids transfer into multiple species. The efficiency of conjugation into enterobacteriaceae patient isolates and a common lab cloning E. coli strain ranged widely from high rates of 10-2 or 10-3 CFU transconjugants/CFU recipients to undetectable, without a clear correlation with the pattern of spread suggested during two hospital outbreaks. In vitro models may not faithfully predict plasmid mobilization until we better understand the most important variables affecting conjugation efficiency.
==== Peptide chain ==== Several of the genes responsible for pyoverdine biosynthesis (e.g., pvdH, pvdA, and pvdF) are involved in the generation of precursor and alternate amino acids necessary for various portions of the molecule. Several others (e.g., pvdI, and pvdJ) are directly responsible for "stitching" together the peptide chain. pvdD terminates the chain and releases the precursor into the cytoplasm, which is consistent with identification of pyoverdine-like molecules in the cytoplasm with incompletely matured chromophores.
Opiates belong to the large biosynthetic group of benzylisoquinoline alkaloids, and are so named because they are naturally occurring alkaloids found in the opium poppy. The major psychoactive opiates are morphine, codeine, and thebaine. Papaverine, noscapine, and approximately 24 other alkaloids are also present in opium but have little to no effect on the human central nervous system. Alkaloids that have no effect on the central nervous system were not always considered to be opiates, but current trend is to refer to all alkaloids derived from opium or poppy straw as such. Very small quantities of hydrocodone and hydromorphone are detected in assays of opium on rare occasions. It appears to be produced by the plant under circumstances and by processes that are not understood at this time. Dihydrocodeine, oxymorphol, oxycodone, oxymorphone, metopon Possibly other derivatives of morphine and/or hydromorphone also are found in trace amounts in opium. Despite morphine being the most medically significant opioid, larger quantities of codeine are consumed medically, most of it synthesized from morphine. Codeine has greater and more predictable oral bioavailability. Codeine is not reliably metabolised into its active form, morphine, by CYP2D6 due to the considerable amount of polymorphism. Many individuals lack any appreciable metabolism to morphine and experience no therapeutic effects, although may still have nausea/vomiting or constipation.

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
