Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.
Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.

Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
=== First term (2018–2019) === Talarico launched his campaign for the Texas House shortly after incumbent state legislator Larry Gonzales chose not to run for reelection. He won the concurrent special and general elections against Republican nominee Cynthia Flores, garnering media attention for walking the full length of the district. He was sworn into the Texas House of Representatives on November 20, 2018, and appointed to the Public Education and Juvenile Justice Committees. In the 86th Texas Legislature, Talarico filed the Whole Student Agenda, a 24-bill public education policy legislative package. The legislature passed two bills from this list. The first, HB 3012, required that students suspended from school have an alternative means to receive coursework. HB 455, also sponsored by Talarico, was written to provide a standardized school recess policy. Governor Greg Abbott vetoed the bill.
Polymer-protein hybrids are a class of nanostructure composed of protein-polymer conjugates (i.e. complexes composed of one protein attached to one or more polymer chains). The protein component generally gives the advantages of biocompatibility and biodegradability, as many proteins are produced naturally by the body and are therefore well tolerated and metabolized. Although proteins are used as targeted therapy drugs, the main limitations—the lack of stability and insufficient circulation times still remain. Therefore, protein-polymer conjugates have been investigated to further enhance pharmacologic behavior and stability. By adjusting the chemical structure of the protein-polymer conjugates, polymer-protein particles with unique structures and functions, such as stimulus responsiveness, enrichment in specific tissue types, and enzyme activity, can be synthesized. Polymer-protein particles have been the focus of much research recently because they possess potential uses including bioseparations, imaging, biosensing, gene and drug delivery.
== Production == In 2009, the worldwide capacity for producing formic acid was 720 thousand tonnes (1.6 billion pounds) per year, roughly equally divided between Europe (350 thousand tonnes or 770 million pounds, mainly in Germany) and Asia (370 thousand tonnes or 820 million pounds, mainly in China) while production was below 1 thousand tonnes or 2.2 million pounds per year in all other continents. It is commercially available in solutions of various concentrations between 85 and 99 w/w %. As of 2009, the largest producers are BASF, Eastman Chemical Company, LC Industrial, and Feicheng Acid Chemicals, with the largest production facilities in Ludwigshafen (200 thousand tonnes or 440 million pounds per year, BASF, Germany), Oulu (105 thousand tonnes or 230 million pounds, Eastman, Finland), Nakhon Pathom (n/a, LC Industrial), and Feicheng (100 thousand tonnes or 220 million pounds, Feicheng, China). 2010 prices ranged from around €650/tonne (equivalent to around $800/tonne) in Western Europe to $1250/tonne in the United States.

The generation of a protein sequence is much easier than the determination of a protein structure. However, the structure of a protein gives much more insight in the function of the protein than its sequence. Therefore, a number of methods for the computational prediction of protein structure from its sequence have been developed. Ab initio prediction methods use just the sequence of the protein. Threading and homology modeling methods can build a 3-D model for a protein of unknown structure from experimental structures of evolutionarily-related proteins, called a protein family. Predictive machine learning-based approaches tackle the structure problem at multiple levels. At the 1D level, secondary structure and solvent accessibility are predicted. The 2D level works on distances and points of contact along the protein chain; these predictions are orientation independent. At the 3D level, the coordinates of all the atoms in the protein are estimated; this level is the primary goal of most prediction efforts. Finally, the 4D level predicts complexes of multiple proteins. Progress at these levels is assessed annually at the biannual Critical Assessment of Structure Prediction event. The results from structure studies can be fed in to machine learning techniques deployed to understand protein-protein interactions.
== Limitations == SELDI is often criticized for its reproducibility due to differences in the mass spectra obtained when using different batches of chip surfaces. While the method has been successful with analyzing low molecular weight proteins, consistent results have not been obtained when analyzing high molecular weight proteins. There also exists a potential for sample bias, as nonspecific absorption matrices favor the binding of analytes with higher abundances in the sample at the expense of less abundant analytes. While SELDI-TOF-MS has detection limits in the femtomolar range, the baseline signal in the spectra varies and noise due to the matrix is maximal below 2000 Da, with Ciphergen Biosystems suggesting to ignore spectral peaks below 2000 Da.
A ubiquitin ligase (also called an E3 ubiquitin ligase) is a protein that recruits an E2 ubiquitin-conjugating enzyme that has been loaded with ubiquitin, recognizes a protein substrate, and assists or directly catalyzes the transfer of ubiquitin from the E2 to the protein substrate. In simple and more general terms, the ligase enables movement of ubiquitin from a ubiquitin carrier to another protein (the substrate) by some mechanism. The ubiquitin, once it reaches its destination, ends up being attached by an isopeptide bond to a lysine residue, which is part of the target protein. E3 ligases interact with both the target protein and the E2 enzyme, and so impart substrate specificity to the E2. Commonly, E3s polyubiquitinate their substrate with Lys48-linked chains of ubiquitin, targeting the substrate for destruction by the proteasome. However, many other types of linkages are possible and alter a protein's activity, interactions, or localization. Ubiquitination by E3 ligases regulates diverse areas such as cell trafficking, DNA repair, and signaling and is of profound importance in cell biology. E3 ligases are also key players in cell cycle control, mediating the degradation of cyclins, as well as cyclin dependent kinase inhibitor proteins. The human genome encodes over 600 putative E3 ligases, allowing for tremendous diversity in substrates. Certain E3 ligases have been utilized in targeted protein degradation applications.

=== Infertility === It can be difficult to become pregnant with PMOS due to irregular ovulation. The first management step is to improve the general health of the mother, such as through lifestyle interventions. GLP-1 agonist for weight loss can be used, but should be stopped at least 2 months before conception. As of 2025, there is not enough data to know if they are safe in pregnancy. Pregnancy in PMOS is more risky than normal, and treatment is focused on getting a single pregnancy, rather than, for instance, twins (multiple pregnancy). With treatment, the eventual family size of women with PMOS does not seem different to those without. The first-line medical treatment for infertility in women with PMOS is letrozole (Femara) to induce ovulation. This is, in general, more effective than clomiphene citrate to improve both pregnancy rates and live births. Other medications that can be used to treat infertility, listed from most to least effective, are metformin + clomiphene citrate, clomiphene citrate alone, and metformin alone. Women may be more likely to experience gastrointestinal side effects with metformin. Gonadotrophin therapy may be effective too, but requires monitoring and increases the risks of multiple pregnancies. When medication and lifestyle interventions are ineffective, infertility can be treated with a laparoscopic procedure called "ovarian drilling", which involves puncture of 4–10 small follicles with electrocautery, laser, or biopsy needles.
==== Keeping the tradition alive ==== Family members often gather to cook lefse as a group effort because the process is more enjoyable as a traditional holiday activity. This gathering also provides training to younger generations keeping the tradition alive. The Sons of Norway have lodges to teach making lefse to younger generations. A lodge in Vancouver, Washington, uses up to 60 pounds (27 kg) of potatoes to make lefse every month. While lefse used to be eaten as a snack food, it is now more often made in large quantities for such lutefisk dinners. Some professional lefse makers can roll 85 to 100 lefse per hour, or even up to 140.
== Business == The company manufactures a variety of GPS (Global Positioning System) trackers intended for use in vehicles and other mobile assets. It also offers an Internet-based subscription service that gives clients access to 5- or 10-second updates around the clock from each installed GPS tracker. The system collects and stores data from each unit on a 24/7 basis, and can generate reports showing the activity of each vehicle during a specified time frame. The system can be configured to transmit alerts to the account owner if certain user-defined conditions are detected, such as a monitored vehicle exceeding acceptable speeds or entering/exiting a specific area.

Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
Oxidation mainly affects methionine residues and is promoted by dissolved oxygen, trace transition metals, and prolonged exposure to light. Buffer choice and the presence of antioxidants in a formulation can alter the rate appreciably. Because the products differ in mass by only a few units, mass spectrometry is often required to detect them.
Published data on long-term ambient stability are limited, so the question remains open rather than settled. Short excursions during transport are common in practice, and many suppliers use insulated packaging with cold packs. Where stability data are absent, cold-chain handling with temperature logging is the safer approach.
Lower temperatures slow hydrolysis, oxidation and aggregation reactions that degrade the molecule over time. Lyophilised powder is more tolerant than solution, but both benefit from controlled conditions.
